Improving in vitro regeneration system in Nicotiana tabacum L. (tobacco)

Abstract (EN)

In this research, Nicotiana tabacum L. cv. Samsun regeneration system has been improved in in vitro conditions. Organogenesis were realized from different explants of Nicotiana tabacum L. Samsun. Organogenesis occured directly from leaf explants and indirectly from stem explants. Callus induced from leaf explants, on MS (Murashige and Skoog, 1962) medium supplemented with different concentration of plant growth regulators. When we compared different callus induction medium, best callus induced medium was included 2,0 mg/L NAA and 0,2 mg/L BAP. Callus in optimum medium subcultured and then regenerated to whole plant on MS medium with 2,0 mg/L BAP, 0,5 mg/L NAA. Shoots formed from stem explants on MS medium with 2,0 mg/L NAA and 0,2 mg/L BAP. Leaf explants formed 2,05 shoots per explants and stem explants formed 7 shoots per explants. Whole plants, which occured directly or indirectly organogenesis, formed roots on basal MS medium. After a while in vitro regenerated plants transplated to soil and acclimizated to ex vivo. At the end of the research we claime that all regenerated plants adaptated ex vivo conditions and growed healthy.
