Abstract (EN)

Aim; Our aim in this study is to investigate the total antioxidant status (TAS), total oxidant status (TOS) and Oxidative Stress Index (OSI) values of diclofenac sodium, which is a non-steroidal anti-inflammatory analgesic, after sciatic nerve injury, to examine the histopathological changes on the sciatic nerve, and to examine the results and enable important changes in treatment. Objective: For adaptation to the environment, the animals were placed in the experimental environment 1 week before hand, and fed with standard rat chow at 20-22 degrees Celsius in an environment of approximately 55% humidity, 12 hours in light and 12 hours in darkness, with optional tap water. The study was divided into 5 main groups as control, sham, trauma, isotonic fluid and diclofenac sodium. Sham, trauma, isotonic fluid and diclofenac sodium groups were divided into four subgroups as 1st day, 3rd day, 7th day and 14th day. Considering the statistical significance of possible losses, each group was formed from 7 rats. Except for the control group, the skin and subcutaneous tissue were passed through a linear incision just below the os femoralis after povidone iodine antiseptic on the right side in the prone position, and the sciatic nerve was reached after blunt dissection of the m.biceps femoralis. Sham groups only underwent surgery. Trauma groups received only injection damage. 0.5 ml of isotonic fluid was injected into the sciatic nerve to the isotonic fluid groups. Diclofenac groups were injected with 2mg/kg/0.5ml diclofenac sodium into the sciatic nerve. All subgroups were sacrificed on day 1, day 3, day 7 and day 14, and blood and nerve tissue samples were taken. Subjects in each group were evaluated daily by neurological examination according to the Tarlow scale. Xylasine (30mg/kg) and ketamine hydrochloride (200mg/kg) were used intraperitoneally for sacrification. Findings: Statistically significant differences were found between control, sham, trauma, isotonic and diclofenac groups in biochemical TAS, TOS, OSI measurements and histopathological examination and immunostaining findings. Conclusion: In our study, it was determined that diclofenac sodium causes pathohistological and biochemical changes and loss of function in neural tissue damage. In order to reduce the risk of post-injection neuropathy, it is important to prefer other drug applications rather than IM drug applications as much as possible, and if necessary, the ventrogluteal region must preferred. In addition, allied health workers who administer drugs should be trained in this regard and personal anatomical variations should be taken into account.
