Analysis of LPIN 2 and SIRT 6 Gene expression in different tissues of Sitagliptin treatment in obesity

Obesity ranks fifth among the most risky diseases. It is linked to impaired glucose tolerance due to type 2 diabetes and insulin resistance; Damage to beta cells in the pancreas can lead to insulin deficiency, which affects glucose used by skeletal muscle, liver, and adipose tissues. LPIN 2 expression is increased in the liver of mice with an obese model due to a high-fat diet. Overexpression of LPIN 2 impairs insulin signaling in the liver. SIRT 6, on the other hand, influences cellular homeostasis by regulating DNA repair, telomere maintenance, glucose and lipid metabolism, thereby influencing diseases such as diabetes, obesity, heart disease, cancer, as well as many metabolic pathways associated with aging, including infection. In our study, it was aimed to develop different treatment methods by using sitagliptin, which is used in Type 2 diabetes, in the treatment of obesity by creating experimental animal models. Of 32 wistar albino rats, 16 of them were fed with normal chow and 8 of them were given sitagliptin, 16 of them were obese on a high-fat diet and 8 of the obese rats were given sitagliptin. Animals were divided into Control, Control + Sitagliptin, Obese, Obese + Sitagliptin groups. The mRNA levels of LPIN 2 and SIRT 6 genes in the liver, muscle and adipose tissues of 24 of 32 wistar albino rats were measured by RT-PCR method and statistically evaluated
