In this thesis study, Culex pipiens complex species populations that distributed in the Central and Eastern Black Sea Region were sampled in order to determine the molecular nature of the insecticide resistance. DNA isolation was carried out after morphologically identification of the species which were collected. The resistance status was evaluated according to kdr L1014F, ace-1 F290V (Agarose gel electrophoresis image and sequencing), ace-1 G119S (sequencing) mutations at the end of the PCR. The presence of homozygous resistant, homozygous sensitive and heterozygote resistant individuals for the kdr L104F locus was observed. The heterozygosity was also high as confirmed by the sequencing results with regional variation. It was observed that the frequency of the ace-1 gene F290V mutation was very low throughout the region. This result confirmed both by gel electrophoresis and sequencing, and there was no homozygous resistant individual in the field. In the ace-1 gene G119S mutation region, the frequency of homozygous resistant individuals were found low level and the proportion of heterozygotes were found higher than the F290V mutation. The presence of in all three mutations was determined throughout the study region, but the rate of kdr mutation was found to be higher than that of ace mutations. This confirms the existing molecular resistance to both the pyrethroids and the organophosphorus / carbamate groups. The mutation types observed in all three regions showed that the selection pressure was high on pyrethroids and low on organophosphorus/carbamate groups.
