Comparative evaluation of the effects of ozone, diode laser and traditional cavity disinfectants on mikroleakage

Abstract (EN)

Aim:The aim of this study was to investigate in-vitro effect of ozone application, laser irradiation and traditional cavity disinfectants on microleakage. MaterialandMethods:In this study,90 freshly extracted human permanent third molar teeth, were used. After extraction soft tissues on teeth were cleaned with a scaler and polished with brush and pumice. Until the date use of teeth were stored in distilled water at room temperature. Standard class V cavities were prepared on buccal surface of the teeth with cylindirical diamond burs under water cooling. Each cavity were prepared as 3mm mesio-distal diameter, 2mm occluso-gingival diameter and 1,5 mm depth. Enamel margin of the cavities were bewelled in a 0,5-1 mm area. After wards the teeth were randomly assigned to six groups (n:15). Group 1: benzalkonium chloride, group 2:chlorheksidine gluconate, group 3: sodium hypochlorite, group 4: diode laser, group 5: ozone. Group 6: control group, will not treated with any application. The primer and bond (Clearfil protect bond, Kuraray Medical Inc. Japan) were applied to prepared cavities respectively according to manufacturers? recommendations and were polymerized by LED (Light Emitting Diode-Elipar Freelight, 3M ESPE, Germany) device for 10 seconds. A hybrit composite resin restorative material (Clearfil AP-X, Kuraray Medical Inc, Japan) was applied in cavities and was cured for 20 seconds by LED device. Before finishing all specimens were immersed into incubator at 37 oC for 24 h. After 24 h, polishing and finishing of restorations were done by diamond finishing burs under water and were completed with aluminium oxide coated disks (Sof-Lex, 3M ESPE, St. Pau, MN, USA). After waiting in the incubator at 37 oC for 24 h, teeth were subjected to thermocycling 1.000 times with a dwell time of 30 second at 5 ± 2 ºC and 55 ± 2 ºC. Then, root tips of samples was closed with resin composite and all teeth, except 1mm surrounding of the restorations, were coated twice with acid resistant varnish and stained with 0.5 % basic fuchsine at 37oC incubator for 24 h. The teeth were sectioned bucco-palatinally/lingually and, microleakage scores of occlusal and gingival walls were evaluated with a standardized scala from 0 to 4 under stereo microscope at 15X magnification. A tooth from each group were selected randomly. The teeth were covered with Pt (platinium) in ion plating unit (BAL-TEC SCD 050, Capovani Brothers Inc., Scotia, NY, USA). And also SEM (Scanning Electron Microscopy) (LEO EVO 40, LEO Ltd., Cambridge, UK) photographs of resin-tooth hard tissue interfaces were taken at different magnifications after microleakage assessment. Results of microleakage test were statistically analyzed by Kruskall-Wallis. Results :In terms of gingival and occlusal microleakage among groups, differences were not determined significant (p>0.05). Occlusal microleakage scores of ozone were observed considerably lower than other 5 groups (p>0.05). Also gingival microleakage scores of ozone were lower than other groups. Conclusion : Statistically there was no difference among groups (p>0.05). However, differences were found among the avarages of groups. The lowest microleakage score were found in ozone group. On the other hand the highest mikrolaekage scores were found in control group.
