Abstract (EN)
PANDAS (Pediatric Autoimmune Neuropsychiatric Disorderders Associated with Streptococcal Infections) is an newly defined ilness in neuropsychiatry. This ilness occurs with a otoimmune mechanism after a Grup A Beta Haemolytic Streptococcus infection. Streptococcus infection causes obsessive compulsive disorder or tic disorders and also indirectly causes PANDAS. In the patient?s immune system, firstly B lymphocytes susceptible to M group of antigens, kils them. After B lymphocyt activation, some of the susceptible B lymphocytes pass over blood-brain barrier and associates with the cell membrane of neuron cells because of epitope mimicry. Outcome of this association neurons are killed by the patient?s apoptosis mechanism. The loss of neurons in the part of the brain which is supposed for coordination of moving and care and also active memory cannot be compensation and neuropshychiatric signs are un avioidableTumour necrosis factor is an important cytokine and also have important role in the apoptosis mechanism of autoimmune diseases. It is expressed by TNF-? gene. The aim of our study was to investigate if there is relationship between -308 G/A and -850 C/T polymorphisms that are in promoter region of the gene and PANDAS disease. We took 3-4 cc of patient?s blood to the tube which involves EDTA. We isolated DNA of the patients. Areas where polymorphisms located in, was amplificated with Polymerase Chain Reaction (PCR) method. Polymorphisms were determined by using available restriction enzymes with Restriction Fragment Length Polymorphism (RFLP) and PAGE (Polyacrylamid Gel Electrophoresis) method. Present study was progressed with control group. In this study, we observed positive significant difference between -308 G/A and -850 C/T polymorphisms and PANDAS.
