Abstract (EN)
In this study, 56 female Wistar albino rats weighing 250-300g were used. Rats; control group, 250 mg / kg dexpanthenol group, 500 mg / kg dexpanthenol group, 60mg / kg tocopherol group, toxicity group, 250 mg / kg dexpantenol + 60 mg / kg tocopherol group, 500 mg / kg dexpanthenol + 60 mg / kg tocopherol group were divided into 7 groups (n=8). Sham group was treated with saline intramuscularly and intraperitoneally for 7 days. On the 8th day liver and blood samples were taken for histological and biochemical examination. No toxicity treatment was used. First treatment group received 250 mg / kg dexpanthenol intraperitoneally for 7 days. At the end of the 7th day, rats left open for 24 hours were given 2000mg / kg paracetamol for toxicity by oral gavage method. The second treatment group received 500 mg / kg dexpanthenol intraperitoneally for 7 days. At the end of the 7th day, fasting rats 24 hours before, 2000 mg / kg paracetamol was administered by oral gavage to create liver toxicity. The third treatment group received 60 mg / kg tocopherol intraperitoneally for 7 days. At the end of the 7th day, fasting rats 24 hours before, 2000 mg / kg paracetamol was administered by oral gavage to create liver toxicity. In the 4th treatment group, 60 mg / kg tocopherol and 250 mg / kg dexpanthenol were given for 7 days. At the end of the 7th day, fasting rats 24 hours before, 2000 mg / kg paracetamol was administered by oral gavage method to create liver toxicity. In the 5th treatment group, 60 mg / kg tocopherol and 500 mg / kg dexpanthenol were administered intramuscularly and intraperitoneally for 7 days. At the end of the 7th day, fasting rats 24 hours before, 2000 mg / kg paracetamol was administered by oral gavage method to create liver toxicity. İn the toxicity group was given 2000 mg/kğ paracetamol by oral gavage method to create liver toxicity. 24 hours after toxicity, rats were sacrificed and blood and tissue samples were made for histological and biochemical examination. As a result of biochemical examination, tissue MDA and MPO values were significantly increased in the treatment groups compared to the control group (p<0.05). It was significantly lower than the toxicity group (p<0.05). Tissue GSH Px levels of treatment groups were again significant compared to control group (p<0.05). Serum ALT, AST and LDH levels were significantly higher in the treatment groups compared to the control group; significantly lower than the toxicity group (p<0.05). No statistically significant difference was found between high and low doses among the treatment groups (p>0.05). Histopathological evaluation of dexpanthenol and tocopherol groups; less damage compared to toxicity group.
