The value of immunofluorescence mosaic based biochip method in autoimmune bullous diseases out of pemphigus

Abstract (EN)
Autoimmune bullous diseases are a group of diseases rarely seen, characterized by autoantibodies against structural proteins of the epidermis or the dermal-epidermal junction. The diagnosis is based on generally the combination of clinical features, histologic and immunofluorescence findings and/or ELISA test. In this study, we aimed to state the value of a new method in the diagnosis of non-pemphigus autoimmune bullous diseases, by using EUROIMMUN biochip mosaic-based indirect immunofluorescence technique. In this study 20 BP anda 5 EEB, in total 25 patients in active phase of the disease and 20 healthy controls who admitted to Dermatology and Venereology Outpatient Clinic of Akdeniz University Faculty of Medicine were included. The patients ABSIS scores, immunflourescence finding, Tip VII Collagen and antiBP180 autoantibody titers by ELİSA (EUROIMMUN®, Lubeck, Germany) test were recorded. All patients and controls sera were tested by EUROIMMUN biochip mosaic-based indirect immunofluorescence technique. Biochip technique compared with gold standart diagnostic tests, sensitivity and spesifisity were calculated for validity. The sensitivity and specificity of biochip technique in the diagnosis of BP was found %90 and %95. In detecting anti-BP180 autoantibodies, a good rate of agreement was observed among biochip technique and ELISA test (p<0,01). The BİOCHİP method of EBA diagnosis may be a suitable method. However, because of the low number of patients in our study, evaluation sensitivity and specificity analysis were not performed. As a result; The BIOCHIP method can be used as a screening test in the diagnosis of autoimmune bullous diseaes and differential diagnosis as a less invasive method which is cheaper, practical, less time-consuming and possible with high sensitivity and specificity in BP diagnosis. In the diagnosis of EEB, the BIOCHIP method seems to be a good diagnostic tool. There is a need to study a wider sera test.
