Investigation of antioxidant and cytotoxic effects on some cancer cell lines of pistacia vera extracts

Abstract (EN)

Pistacia vera is grown widely for commercial purposes in the Mediterranean and southern regions of Turkey. The aim of this thesis is to investigate the usability of P.vera skins, which is an industrial-waste, as antioxidants in the food-industry and anti-carcinogenic substances in the health sector. Therefore, the flavonols, flavon, flavan-3-ol, flavanone, and phenolic acids extracts prepared by P. vera fruit shells from Antep, Siirt and Urfa regions were determined and identified prominent components; gallic, protocatechuic, 4-hydroxy-benzoic, benzoic acid. The IC50 values of DPPH and hydroxyl radical scavenging capacities of P. vera extracts were in the range of 2,08-47,99 and 4,93-32,83 ppm, respectively. When the metal chelating and reducing potentials of P. vera shell extracts were evaluated, IC50 values were determined in the range of 0,453-163,638 mg/ml and 148,38-9,41 ppm-Vit-CEq respectively. Anti-proliferative effects of phenolic component extracts of P. vera were investigated for HeLa, MCF-7, OE-33 and ACC-201 cancer cell lines. The best IC50 values for each cell lines were obtained with -Antep-flavanone (25ppm), -base and then acid hydrolyzed of Urfa solid phase (25ppm), -base hydrolyzed of Antep-Urfa solid phase(45ppm) and -acid hydrolyzed extracts of Siirt solid phase (28 ppm), respectively, at 24 hours of incubation.To the results, P.vera outer shell is an important source for gallic acid, which is one of the phenolic components, and all antioxidant properties can be defined as good when compared with the literature data; cancer cell lines HeLa, MCF-7 and ACC-201, as well as a particularly resistant cell line OE-33 can be mentioned as remarkable features.
