In this thesis; gene encoding lactate dehydrogenasefrom Plasmodium vivax was amplified by adding 6 histidines to the C-terminal of the enzymeprior to expressıon of the protein in the expressıon vector pKK223-3, in E. Coli. Theoverproduced protein was then purified by using Ni-NTA Spin kit and Ni-NTA agarose andanalysed kinetically. An antibody produced against PfLDH was tested against this kineticallyanalysed pure PvLDH protein. It was shown that two proteins were similar enough that PvLDHreacted with PfLDH antibody. It is expected that the drug design studies applied on PfLDH arealso made applicable on PvLDH after obtaining pure PvLDH protein. As a result it is expectedthat, if we develop one antimalarial against one at the Plasmodium species would work againstother species too.Keywords: Malaria, Plasmodium vivax, Lactate dehydrogenase, Expression, Purification.
