Synthesis and characterization of poly(1-azabicyclo[4.2.0]octane) polymers and their derivatives

Abstract (EN)
In recent years there have been made so many research which are intended for developing functional biopolymer systems. In most of the studies until now, it is emphasized on the synthesis, characterization and the diversity of biologically featured polyelectrolytes. In this thesis, the synthesis of a biodegredable polyelctrolyte, (polyconidine) which has an heteroatom in its structure is explained. For poly(1-azobicyclo[4.2.0]octane) obtaining, initially the synthesis of the monomer, 1-azobicyclo[4.2.0]octane (conidine) is carried out by organic methods which then will be used in polymer synthesis. Thereafter, polymers having different molecular weights are synthesized and the characterization of these polymers are done by different methods such as UV, FT-IR, SEC and Ultracenrifuge. By the modification of obtained polymers with bromoacetic acid, gaining some suitable physicochemical properties to polymer is studied. FT-IR and UV spectra of substances formed on each step of conidine synthesis were recorded and the pathway of the synthesis was checked constantly. Synthesized conidine was used as monomer and in the existance of different initiators (methyl iodide, ethyl bromide, hexadecyl bromide, t-butyl bromide, benzoyl chloride and trimethylsilane bromide) poly(1- azobicyclo[4.2.0]octane) was obtained by cationic ring opening polymerization. The effect of changing Initiator/monomer ratio on the polymers? characteristics was also examined. For the chemical modification of poly(1-azobicyclo[4.2.0]octane), bromoacetic acid is used and polyampholyte synthesis is achieved with the quaternization of the polymer. It is known that the physicochemical properties of polyelectrolytes is directly related to the structure. For this reason the characterization of synthesized poly(1-azobicyclo[4.2.0]octane) is intended. FT-IR spectra of poly(1-azobicyclo[4.2.0]octane) is recorded and molecular weights, Mw/Mn values, Mark-Houwink constants and molecular diameters of the polymers are measured with size-exclusion chromatography with four-detectors (light-scattering, refractive index, viscosity and UV). With Zeta Sizer, size-analysis of polymer chains are done and their zeta potentials are measured. Svedberg constants that belongs to the obtained polymers and the graphics for sedimentation values are determined with ultracentrifuge. It is determined that molecular weights of the polymers are affected by the change in the amount and the type of initiators used and also from the polymerization media. Analysing of having biodegradable characteristics of the synthesized polymers has been being reviewed. The effect to the cell proliferation of polymer derivatives with bromoacetic acid was observed with breast cancer cells (MCF 7) in vitro enviroment. The effect to the cell proliferation of polymer concentration was observed. Key words: polyelectrolyte, policonidine, cationic ring-opening polymerization
