Abstract (EN)

Polyporaceae family members especially Ganoderma lucidum and Trametes versicolors are important for their medical properties. In this study for the first step Ganoderma lucidum (Turkish basidiomycetes isolate) is being searched using extensive cultivation systems adapted the traditional growing methods developed in Japan and China. This research also focuses on indoor techniques which permit better environmental control for the isolate.The second step, molecular data analyses of four basidiomycete isolate are studied using molecular phylogenetic techniques by the nucleotide sequences of ITS-5.8S regions of rDNA to establish the basic taxonomy of the isolates. The analysis resulted in the emergence of four major clades. Two of the clades belonged to the Ganoderma and similar to Ganoderma lucidum, and Ganoderma applanatum the third clade was belonged to Trametes and similar Trametes versicolor and finally the fourth clade was belonging to the Trametes sp.. but similarity for the subgenus could not recognized.For the third step interspesific hybridization between G. lucidum and G. applanatum, and Trametes versicolor and Trametes sp.. were studied using protoplast technique. To establish conditions for the protoplast isolation and regeneration of the mycelia of the isolates, various enzymes and osmatic stabilizers were examined. Among different media MCMM medium was found to be the best for colony growth. Lyticase was the best lytic enzyme and mannitol was the best osmatic stabilizer with the fusion frequency in the range of 0.030 %. Protoplasts from various hybrids were released and regenerated to determine whether they were heterokaryons. In all regenerated colonies, both parental types could not be recovered at the same time. For being sure for the fourth step isozyme analysis up to esterase isozyme was determined with the parental isolates and fusants. The electroforetic bands of the (Ganoderma sp.. and Trametes sp..) fusants showed whether one or two parental esterase bands. KeyWords Polyporaceae, Ganoderma lucidum, protoplast, ITS, 5.8 S
