Abstract (EN)

SUMMARYCarbonic anhydrase (E.C.4.2.1.1) is a metalloenzymes contains Zn+2 ion. In human erythrocyte carbonic anhydrase enzyme catalyze the reactions of CO2 hydration in cells or the dehydration reactions of HCO3?s. Carbonic anhydrase shows activity in kidney, gastric mucosa and the lens of the eye, salivary glands, brain, nerve myelin sheath,pansreas, prostate, uterus and endometrial tissues. In addition, it is well known that this enzyme has an important role on endocrine organs and gills of fish, some insects and bacteria, making the shell of crustaceans and the formation of the egg shell, algae and photosynthetic chloroplast of plants.The Carassius gibelio used in this study is healthy and mature. The length of Carassius gibelio is 35 cm and the average weight is 3000 grams. They live in parts of the muddy bottoms of fresh water. They can survive even in water contains very low amount of oxygen. The average water temperature has been established between 10-20 oC. Carassius gibelio is invasive spcies and also they do not have economic value. It has been tried to determine that optimum pH, stable pH, optimum temperature and ionic strength of homogenates which gained from this fish. The optimum temperature, pH and ionic strength of Carassius gibelio has been found as 25 oC, 8,7 and 1 M respectively the of, the optimum pH, ionic strength the. Afterwards, carbonic anhydrase enzyme in gills has been tried to be purified by using affinity chromatography (Sepharose- 4B-tyrosine-sülfonilamid).Key words : Carassius gibelio, Sepharose-4B-L-tyrosine-sülfonilamid, optimum pH, stable pH, ionic strength.
