Abstract (EN)
Biofilm is a polysaccharide matrix structure formed by bacteria on biotic and abiotic surfaces, and it is a layer that allows the bacterial cell to protect itself against external factors, thereby continuing to reproduce itself by resisting various disinfectants. Hospital infections, infection of artificial materials found in the body, biofilm formation, which is seen as the cause of many infections threatening food safety, are controlled by the 3',5'-monophosphate (c-di-GMP) nucleotide in the bacterial cell. The most important feature of c-di-GMP is that it can be attached to different molecules in different ways by having different conformations. Thanks to its guanine base, such as DNA and RNA structure, the c-di-GMP molecule can also form a quad lattice structure called G-quadruplex. Various small molecules attach to this cage structure and inhibit c-di-GMP activity. Structures such as proflavin, acriflavin, acridine, and anthracene, which contain multiple aromatic rings, can usually enter the G-quadruplex structure, thereby stopping bacterial functions controlled by c-di-GMP. Similarly, some heterocyclic compounds containing aromatic rings are thought to have the potential to exhibit such a property. In organic chemistry, heterocyclic compounds are a very large family and of great importance. According to the literature, a significant part of many compounds and drug active substances occurring in nature are heterocyclic compounds. There are many different methods for the synthesis of such compounds. One of the most effective and advantageous methods is carben transfer reactions of diazo compounds in the presence of metal catalysts. Depending on the type of catalyst used and reaction conditions, the synthesis of different heterocyclic compounds is possible by using conjugated carbonyl compounds as the substrate. In this thesis study, rhodium acetate or copper acetyl acetonate catalyzed reactions of ethyl diazoacetate with different α,β-conjugated amides having different functional groups and suitable conformations were examined and reaction products were evaluated The aim of the study is to understand which compound is preferred depending on different functional groups and different conditions in the synthesized amide compound, to characterize the products formed and to obtain compounds with high reactivity, which were not previously included in the literature.
