Effect of POU1F1 genotypes on growth traits in kil goats

Abstract (EN)

Objective: In this study performed in Kıl goat kids, it was aimed to identify AluI and PstI polymorphisms in the 450 base pair long sequence consisting of intron 5, exon 6 and 3' flanking regions of the POU1F1 gene and to reveal the differences between POU1F1 genotypes in terms of kid growth characteristics under breeder conditions. Material and Methods: The study was carried out on 150 Kıl goat kids from 6 different farms in the villages of Aydın and Denizli provinces. In order to determine the growth characteristics of the kids, the birth weight, the body weights at the ages of about 3 and 6 months, and the average daily live weight gains from birth to the age of 3 and 6 months were determined. With the primer pair designed specifically for the POU1F1 gene region, 450 bp PCR products were successfully obtained in 138 of 150 samples. In order to reveal the nucleotide polymorphism in the amplified target DNA region, PCR products were cut separately with AluI and PstI restriction enzymes. The PCR-RFLP product DNA fragments formed after enzyme digestion were separated according to their size in gel electrophoresis and visualized. Results: As a result of the analysis of the genotype data obtained from the images, the frequencies of the TT, TC and CC genotypes obtained by the POU1F1/AluI restriction were found to be 0,86, 0,13 and 0,01, respectively, and as a result, the frequencies of the T and C alleles were 0,93 and 0,07, respectively. The frequencies of the TT and TC genotypes determined by the POU1F1/PstI restriction were found to be 0,95 and 0,05 respectively, while the frequencies of the T and C alleles were 0,97 and 0,03. The CC genotype was not observed in both enzyme cuts and the frequency of the C allele was observed to be very low. Chi-square tests for genotypes obtained by both enzyme cuts revealed that the studied population was in Hardy-Weinberg equilibrium. The least squares means for the birth weights of the kids, their liveweights at the age of about 3 months (average 95,2 days) and 6 months (average 172,7 days), and the average daily body weight gains from birth to 3 and 6 months of age were 2,92 kg, 17,91 kg, 26,53 kg, 156,70 g and 126,91 g, , respectively. The differences between the averages of the genotypes obtained with both POU1F1/AluI and POU1F1/PstI restrictions were found to be statistically insignificant in terms of the growth characteristics examined (p>0.05). Conclusion: In this study, the polymorphism of the examined 450 base pair region of the POU1F1 gene in terms of AluI restriction enzyme in Kıl goats was defined for the first time, and again for the first time, AluI and PstI polymorphisms were associated with kid growth characteristics. In this study, it was defined for the first time that the examined 450 base pair region of the POU1F1 gene exhibited polymorphism in Kıl goats in terms of AluI and PstI restriction enzymes. In the analyzes performed for the data on growth characteristics of the kids, it was determined that the genotypes obtained for the each of two enzyme cuts within the scope of the examined gene region did not make a significant difference in terms of growth characteristics.
