Abstract (EN)
Introduction: Preeclampsia is a specific disease that increase perinatal mortality and morbidity and closely associated with systemic inflammatory response. However the studies which try to determine which one of the triggering reason of inflammation or inflammatory determiner which is more influential on preeclampsia inflammation have continued. The essential factor is still unknown. Three parameters were included in the study regarded as potential risk factors. Milk Fat Globul Epidermal Growth Factor-8 (MFG-E8); is an endometrial epithelial proteins and regulates inflammation and apoptosis. MFG-E8 is an important in the development of endometrium and chorionic villus. Osteoprotegerin (OPG); is pro-angiogenic factor, it can be important for plasental angiogenesis. Suppressor of cytokine signaling-3 Receptor (SOCS-3) has a role on prevents the inflammation. Objective: The possible role of these factors in the etiology of preeclampsia and inflammatory process or aggravation of this situation were asked to investigate. Material and Method: In this study, blood and placental tissue samples were used as materials. Control group is composed of 50 healthy women volunteers with no history of preeclampsia and 28-34th weeks of pregnancy. The experimental group consisted 40 preeclamptic women in 28-34th week of pregnancy. 5 cc blood and tissue samples are taken from preeclamptic group and healthy individuals. Homogenate is prepared from the placental tissue samples which had taken. In order to remove excessive blood, it was conserved for one night in ≤-20 ºC and in 20 ml 1xPBS. It was frozen for second time and enabled the fragmentation of cellular membranes. Homogenates are centrifuged at 5000xg for 5 minutes, the supernatant part was separated and conserved at -20ºC≤ until the working time. All of the measurement analyzed through the ELISA (Enzyme-Linked Immuno Sorbent Analysis) method and findings evaluated comparatively. Statistical analysis was evaluated using SPSS 15.0 version. Results: When analyzed the MFG-E8, SOCS-3 and OPG levels in the serum, there was a significant difference between patient group and control group in OPG levels. In the preeclamptic group, it is observed that OPG levels raised and displayed statistically significant difference with respect to control group (p<0.01). In respect to the MFG-E8 and SOCS-3 it is not observed any difference. When analyzed in the placental tissue; it is found that MFG-E8 tissue levels are statistically significantly low in preeclamptic group with respect to healthy control group and that there is no changes in the levels of (p<0.01) SOCS- Conclusion: MFG-8 is a parameter has not been previously studied and determined low levels of preeclamtic plasental tissue in our study. These results showed that MFGE-8 could be control both antiinflammatory activity or have negative effect on development of placental villi. Serum OPG level may be considered directive on inflammation of this diseases. All these parameters help to monitor the response to tissue. Key Words: Preeclampsia, MFG-E8, OPG, SOCS-3.
