Evaluation of 20q TERRA expression levels in preeclamptic human placenta

Abstract (EN)
Inrtoduction: Telomeric repeats are known to be responsible for chromosome integrity preservation and restoration mechanism. TERRA transcripts are responsible for the conservation of telomeric margins. In preeclampsia, in some studies, telomere length is found to be shortered longitudinally. We anticipated that this telomere shortening in preeclampsia may be associated with TERRA transcripts and we have hypothesized that "In preeclamptic human placenta 20q TERRA expression level may undergo variation". In that ground, in preeclamptic human placenta and healthy human placenta we explored the 20q TERRA transcription levels. Material and Methods: Two cohorts were created corresponding to preeclamptic human placenta (n=6) and healthy human placenta-control- (n=6). Samples were immensed with serum physiologic. From each of the samples producing cDNA were transcription levels of 20q TERRA were analyzed via quantitative real time polymerase chain reaction (q RT-PCR) assay. The data investigated by GraphPad. From every sample for the obtawied tissue samples, routine paraffine follow-up was performed. The 5μm sections were cub by using microtome, hematoxylin&eozin staining was done and photographed. Result: In preeclampsia group, 20q TERRA expression level increased compared to the control placenta at mRNA level. Histological observations showed that trophoblastic buds in the preeclamptic human placenta were intensely observed. Conclusion: In preeclampsia cohort, TERRA expression level incrcased in comparison with healthy decreased control group. These results indicate that telomere shortening seen in preeclampsia may be the result of increased TERRA transcription level.
