Abstract (EN)
Obesity, which is associated withh many metabolic diseases, including COVID-19, has become a worldwide epidemic. The ability of probiotics to support gastrointestinal (GI) health has been known for years. Since alternative approaches against obesity can be developed with an intervention to the GI system, the current study's objective is to determine the impacts of the exopolysaccharides (EPS)s isolated from natural products with probiotic characteristics, both alone and in complex with silver nanoparticles (AgNPs) as an anti-obesity biomaterial. First, using a phenol-sulfuric acid technique, the greatest EPS quantity was found in 10 material samples known to have probiotic qualities. Then, the identification of the microorganism in this chosen sample was performed along with the monosaccharide composition and functional groups of this chosen EPS. The average molecular weight distribution of EPS was also investigated. After the EPS-AgNP biosynthesis optimization processes, optical, compositional, and morphological analyzes of EPS-AgNPs were performed. Thus, the effect of EPS-AgNP and EPS on the adipogenesis process was performed on the 3T3 cell line and compared to control AgNO3(aq) by cell culture techniques. The effects of applied agents on adipogenesis were qualitatively observed with Oil Red O painting. Furthermore, the quantity of triacylglycerol (TAG) in cells was examined. The results demonstrated that the highest concentrations of EPS (2.25 mM) and EPS-AgNP (1.25 mM) decreased adipogenesis by 75% and 85%, respectively, in comparison to the control.
