Determination of aflatoxin m1 detoxification capacities in different milk types of probiotic bacteria

Abstract (EN)
In addition to the nutritional value and health benefits of milk, aflatoxin M1 (AFM1) in milk threatens human health. Our study was conducted to examine the potential use of Lactobacillus acidophilus DSM 20079, Lactobacillus rhamnosus GG and Bifidobacterium bifidum DSM 20456 probiotic bacteria in reducing the toxic effect of AFM1 and the ability of these bacteria to bind AFM1 in cow's milk, goat's milk, sheep's milk and PBS media.Cow's milk, goat's milk, sheep's milk, and PBS media were contaminated with AFM1 at a concentration of 100 ng/l. Various working groups were formed by adding 108 cfu/ml density Lactobacillus acidophilus, Lactobacillus rhamnosus and Bifidobacterium bifidum probiotic bacteria to different milk types and PBS media. 2% inulin was added to half of the study groups and incubated for 4 hours at 37˚C. Then it was stored at +4˚C for 1 day and AFM1 levels were analyzed by ELISA kit.In our study, L. acidophilus between 5,11-39,81%, L. rhamnosus between 2,56-39,71%, B. bifidum between 9,64-40,14%, L. acidophilus+B. bifidum between 12,52-38,55%, L. rhamnosus+B. bifidum showed the ability to bind AFM1 between 7,65-42,90%. The probiotic bacteria strain with the highest AFM1 binding ability was determined as L. acidophilus+B. bifidum in PBS, B. bifidum in cow milk, L. acidophilus in goat milk, and L. rhamnosus+B. bifidum in sheep milk. The level of AFM1 binding by working groups was between AFM1 %2,32-12,52 in PBS, %9,08-40,14 in cow's milk, %15,01-38,01 in goat's milk, %32,49-42,90 in sheep's milk. binding level of AFM1 was highest in sheep milk and the lowest in PBS.The level of AFM1 binding varies in milk species of probiotic bacteria. Binding ability of AFM1 among different milk types was determined from highest to lowest in sheep's milk, cow's milk and goat's milk, respectively.
