Determination of cytotoxic effects of azurine protein extract from Pseudomonas aeruginosa isolates on some cancer cell lines

Abstract (EN)

Based on this research From June 2022 to December 2022, collected 150 clinical samples from patients who were attending differences hospitals in Baghdad, Iraq, and isolate and identify P.aeruginosa from various sources by using classical, bacteriological, and Vitek II methods. After final diagnosis of samples, among 130 positive culture samples, only 32.3%% (42/130) isolates belonged to P. aeruginosa, and 67.7% (88/130) isolates belonged to other genera of bacteria. The susceptibility test of bacterial isolates against 11 selected antibiotics was conducted by VITIK 2which indicates that the P. aeruginosa isolates are fully resistance 42 (100%) to 3 of the antibiotics of interest during the current study, particularly Cefazolin, Ceftriaxone and Trimethoprim /Sulfamethoxazole. Meropenem was the most effective antimicrobial agents against P. aeruginosa isolates because P. aeruginosa, was showed high percentage of sensitivity to it 36 (85.71%), followed by Ciprofloxacin 28(66.66%) and Piperacillin, Tazobactam 26 (61.90%). Then, azurine was purified from the isolated strains using ion-exchange chromatography on Sephadex G-25 and G-75. Azurin was purified using ammonium sulfate, and it was then identified at 14 kda by polyacrylamide gel electrophoresis. Using a light microscope , treated cell cancer lines were examined, and detection azurin gene by PCR. Different concentration of azurin have been used to study the effect of cytotoxicityin the breast cancer cell line lung cancer. The results showed the highest rate of inhibition of brest cancer cells at the concentration of 100µg/ml by 61.200 %. In the lung cancer the highest rate inhibition was 47.200 % at the concentration 100 µg/ml. Our study's findings offer optimism for the development and use of novel cancer therapies that aim to overcome the limitations of current cancer therapies, which may also help to totally eradicate tumors while minimizing side effects and adverse impacts
