Abstract (EN)
Objective: The aim of this thesis is to clone the gene encoding the cellulase of P. putida KT2440 which can be used as a biofertilizer. Material and Methods: The cellulase gene of P. putida KT2440 strain was analyzed by in silico tools and the cellulase gene was identified with genome information. Primers for promoter and structural gene regions were designed for cloning of this gene. The cellulase gene of P. putida KT2440 strain was cloned with the primers and recombinant stock cultures were prepared. Result: In this thesis, the targeted cellulase gene regions of P. putida KT2440 strain were amplified by PCR method. The inserts were transformed into competent E. coli cells with recombinant vectors. For the recombinant DNA fragments detected in the screening results, bacterial stock cultures of the relevant cells were established. Conclusion: Recombinant stock cultures were obtained by transforming the DNA region of the cellulase gene into recombinant cell lines. It will be possible to use these recombinant cultures in future research on cellulase enzyme.
