Role of cellular and humoral immunity in psoriasis pathogenesis

Abstract (EN)
Psoriasis is frequently seen, going on flame and remission period, with unknown etiology, generally unthreaten patients life, chronic and benign disease of skin. Different extent and violent of skin lesion are seen in patients. Different clinic forms could be seen in life period of the same patient. Recently knowledge about psoriasis pathogenesis changed due to studies which made about psoriasis. Beforehand psoriasis has been considered an epidermal differentiation disorder with keratinosit hiperproliferation, recently considered as epidermal hiperplazia and hiperproliferation due to focal immun system activation. Chronic sitimulation and communication disorder with other cell of T cell in the skin are main immunologic mechanism in patogenesis. Cause and mechanism of activation of this cells are unknown. İn this study we aimed to investigate the effects of mometazon furoat with calcipotriol on cellular and humoral respons in patients with plague psoriasis20 adult patients with plague psoriasis who apply to Fırat University Dermatology Clinic between March 2006 and March 2007 were included to this study. Clinic symptoms and finding of patients were evaluated and recorded. Blood and tissue samples of patients were taken before and after treatment. SPSS 10.0 software was used for statistical analyseSignificant differences were determined by using immunohistochemistry in subgroup of lymphocytes (CD4+, CD8+ and CD25+) in tissue samples of patients were taken before and after treatment (P<0.05). After treatment a significant decrease in CD4+ and CD4+CD26+ lymphocytes were determined by using flow cytometry (CD3+, CD4+, CD8+, CD4+/CD8+, CD19+, CD5+CD19+, CD5+, CD25+, CD4+CD25+, CD4+ CD26+ ve CD4+CD30+) (P<0.05). Significant differences were not determined in cytokines level (IL-4, IL-10, TNF-? and IFN-? ) before and after treatment. After treatment, increase of TGF-ß was significant statistically (P<0.05As a result our finding support role of immunity, especially cellular immunity in immunopatogenesis of psoriasis. We recommend that focusing on cellular immunity in immunopatogenesis of psoriasis will be useful at later studies.
