Immobilization of pullulanase on different carriers and investigation of some properties

The pullulanase is a member of the ?- amylase family which is a class of the glycosyle hydrolases. In industry, pullulanase is used for many occasions. The saccharification of the starch, the desizing process in textile, the activity of the stain removers which contains starch in detergant industry and channeling the wood pulp in paper industry are some examples. Despite the pullulanase is used in industry very often, the immobilization of the pullulanase is not researched frequently.In this study, the partial purification of pullulanase than the fungus of Hypocrea jecorina was done. For the first time, the pullulanase of H. Jecorina was immobilized by the methods of adsorption by linking to various carriers, cross linking and entrapment. The proper method for the immobilization of pullulanase was chosen. Later on, the optimum pH, the optimum temperature, thermal stability, of Km and Vmax values of the immobilized enzyme were determined.At last, the proper methods for immobilization of the pullulanase were emerged as: the immobilization of chitosan beads which contains PEG 6000 and the adsorption of the PEG 6000 beads. First one is based on entrapment method and the second one is based on adsorption. According to the entrapment method, the characteristics of the immobilized pullulanase are found as the following; the optimum pH is 8, the optimum temperature is 50?C, the Km value is 0,0184 mg glu./ml and the Vmax value is 0,2499 U/ml. According to the second method which is the adsorption, the characteristics of the immobilized pullulanase were found that; the optimum pH is 7, the optimum temperature is 70?C, the Km value is 0,0595 mg glu./ml and the Vmax value is 0,2160 U/ml.
