Abstract (EN)

Direct measurement method does not exist that allows observation of amount of heparin in the blood. Heparin, is a sulfated polysaccharide molecule used for prevention of clots in extracorporeal applications. Under the presented thesis chemicals with immobilized affinity ligand-induced change in the frequency of interaction with in a flow cell to the surface of quartz crystal is targeted from the human plasma heparin measurements. Chemical immobilization methods to be applied with the ligand molecules on the surface of the crystals used for long duration and is aimed to be repeated. Offered as part of the thesis: 1) Electromechanical and Electronic Design 2) the sensor creation 3) heparin measurements from plasma. To measure the frequency of quartz crystals (12 MHz oscillator circuit and frequency counter circuit with LCD indicator) electronic circuit were designed and manufactured. Teflon flow cell were designed and produced. Determination of heparin were held by creating a closed circuit flow of phosphate buffer and plasma under temperature-controlled environment.
