The investigation of methylene blue effects on survival increase of random patern dermal flaps on rats

Abstract (EN)

Methylene blue inhibits the formation of free oxygen radicals by xanthine oxidase inhibition. Also, it can prevent the effects of NO by inhibiting Guanylate cyclase, nitric oxid (NO), endothelial NO synthase (eNOS), and inducible NO synthase (iNOS).This study was made for whether methylen blue effects or not on flap viability. 28 male Sprague Dawley rats weighing between 250-300 gr were used for this study. Rats were allocated equally to 4 groups. In control group, their flaps were raised and then again sutured into their site. 30 minutes before operation, intradermal injection of 1 ml/kg of % 1 methylene blue was given to intradermal group and then continued for 10 days. After intravenous group?s flaps removed, they were again sutured into their site. 1 ml/kg of % 1 methylene blue was daily administrated through tail vein. After 1 ml/kg of % 1 methylene blue intradermally administrated to chemical delay group for 10 days, the flaps were raised and again sutured into their site and methylene blue was given at the same dose for 10 days.Flaps were evaluated after 10 days. Areas of flaps viability were calculated with digital planimeter. Mean flap viability areas were found as 47.84 % for control group, 33.18 % for intradermal group, 46.35 % for intravenous group, 41,63 % for chemical delay group. When methylene blue intradermal group was compared with control group, flap viability was significantly reduced (p<0.05). According to control group, other groups were not found significant.To administrate % 1 low consantration (1 ml/kg) methylene blue can not increase random flaps viability at rats. The group that administrate intradermally flap viability was reduced.Key Words: Methylene blue, random flap, xanthine oxidase inhibitor, guanylate cyclase inhibitor
