Abstract (EN)

Rasagiline mesylate is a drug used in the treatment of Parkinson's disease. There are oral tablet formulations in the Turkish drug market. Due to the oral use, Rasagiline mesylate undergoes first pass effect and its bioavailability decreases. The brain is separated from the bloodstream by a barrier. This barrier prevents many molecules from entering the brain. Solid lipid nanoparticles are frequently used systems in terms of being targeted to the brain due to their nano-sized lipid structures. In brain-centered diseases, the priority is to cross the blood-brain barrier. Nasal application is one of these methods. It is shown as an alternative way to the oral and parenteral route. Advantages of the nasal administration of the drugs are; rapid absorption, eliminate hepatic first pass effect, thus increased bioavailability and provides targeting to the brain. However, systems that applied to the nose have a short residence time due to the mucociliary movements. Mucoadhesive gel systems have been developed to eliminate this problem. In this study, solid lipid nanoparticles containing Rasagiline mesylate were prepared and combined with the thermosensitive mucoadhesive gel system. Characterization studies were carried out. An experimental design was made to determine the components proportions. Flow properties and viscosity studies were carried out for the applicability of the formulation. In vitro release and ex vivo permeation studies was performed, stability of the formulation was investigated. It was determined that the formulation developed was easy to apply. It is thought that first pass effect was eliminated by nasal administration. In addition, it is thought that Rasagiline mesylate can be absorbed from olfactory region and directly pass into the brain.
