Effect of L-NAME on i̇ntestinal dysmotility i̇nduced by acute pancreatitis in rats

Abstract (EN)
Acute pancreatitis is a common disease with a relatively high morbidity and mortality. Translocation of bacteria from the gut into pancreatic necrosis is an important factor in the development of septic complications and mortality in acute pancreatitis. Suggested underlying mechanism is decreased gut motility. Nitric oxide (NO) represents a potential key mediator of the local and systemic manifestations of acute pancreatititis. NO has well described effects on motility and is believed to be the major inhibitory neurotransmitter in the gut. The study aim was to determine the role of inhibition of inducible nitric oxide synthase (iNOS) in gastrointestinal dysmotility during acute pancreatitis. Thirty-eight Wistar Albino rats were studied. Rats were divided four groups. Group I (n=8) received intraperitoeal (İP) normal salin. Group II (n=10) received six ip injections of cerulein (100 μgr/kg). Group III (n=10) received injections of N-nitro-L-arginin metil ester (L-NAME), (5mg/kg, SC). Group IV (n=10) received cerulein with L-NAME. Gastric emptying and intestinal transit were assessed by the phenol red recovery method. Other investigations included measurement of serum amylase and lipase activity and pancreatic histopathological examination. Plasma amylase, lipase and pancreatic histologic score were significantly induced in Group II rats given cerulein compared with those in Group I rats given saline. Gut motility was inhibited in ceerulein given rats as aspected. However decresed motility was not effected by L-NAME. Acute pancreatitis impairs gastrointestinal motility in rats with differences in the pathogenic mechanisms. L-NAME dosen't effect inhibited gut motility.
