Evulating angiogenic factor releasing after ischemia and effect on angiogenesis in cerebral ischemia rat model

Abstract (EN)

Objective: We study angiogenic activity by using the most important angiogenic factor VEGF expression and value of sVEGFR-1 and sVEGFR-2 receptors in blood. For this, we occlude carotid arters of cerebral arters in rats. The aim of this study is to show in repeating AVM, the occlusion of main arter causes revascularisation, not recanalisation.Materials And Methods: In our study we use 18 female Wistar Albino rats, divided into 3 groups randomly, in each group we use 6 rats. All rats were subjected to anesthesia with ketamine hydrochlorideof 35 mg and xylazine of 5 mg dose during blood sampling from tail veins and before surgery. Group 1 (sham): Only middle neck incision done. Group 2: Right carotid arter ocluded by 3.0 silk suture. Group 3: After oclusion of right carotid arter, left carotid arter ocluded by Yaşargil aneurysm clip (Aesculap FE 721 K) for only 10 minutes, then clip taken off. After 10 days from surgery, again blood sampling from tail vein, and then rats sacrificed, their brains have taken out. In brain specimens, VEGF expression examined by immunohistochemical staining methods. In blood samples before and after surgery, Svegfr-1 and Svegfr-2 values examined by Elisa method.Results: Groups in which cerebral arters occluded, VEGF expression increased, after surgery competetive receptor sVEGFR-1 of VEGF value decreased and in angiogenesis sVEGFR-2 value incresed but the effect mecanism of this is not clear.Conclusions: After occlusion of main cerebral arters, angiogenic factors stimulates angiogenesis in distal arters. By finding evidences of angiogenesis, occlusion of main arters by endovasculer or surgical methods in AVMs is not enough in treatment and the cause of repeated AVMs is revascularisation, not recanalisation. The AVMs can not be treated by embolisation. These are the results of our study.Key Words: Angiogenesis, vascular endotelial grwth factor, arteriovenosus malformation, revascularisation.
