Özet (EN)

Objective: In this study we aimed to investigate the dose-dependent protective effect of hesperidin against to oxidative damage by using biochemical and electron-examination in experimental spinal cord injury in rats. Method: This study was planned as 4 groups and In each group there were 10 Wistar albino rats. G1: laminectomy was performed; G2: laminectomy was performed and extradural clip was applied; G3: laminectomy was performed, extradural clip was applied and low dose hesperidin was given (50 mg/kg); G4: laminectomy was performed, extradural clip was applied and high dose hesperidin was given (100 mg/kg). Thoracal 7-10 laminectomy was performed. Spinal cord injury was created by applying the extradural clip for one minute. 50 mg/kg hesperidin was administered to G3 and 100 mg/kg hesperidin was administered to G4 intraperitoneally after trauma. Rats were sacrificed at the end of 24 hours. Results: In the drug groups (G3, G4), It was no statistically significant decrease on plasma levels of malondialdehyde (MDA) which is the end product of lipid peroxidation and plasma levels of total antioxidant status (TAS) was examined, ıt was no significant differences was observed. When spinal cord samples which were taken after the experimental spinal cord trauma are examined morphologically, a significant reduction in the number of neurons was determined in G2 and G3 groups. High dose hesperidin treated group (G4) compared to control group, the neuron numbers were similar. Conclusion: These findings suggest that Hesperidin can be effective to provide significant improvement on neuronal loss
