The effect of tenoxicam on over ischemia-reperfusion damage in rats

Abstract (EN)

AIM: This study aimed to demonstrate the role of tenoxicam (TNX) in the prevention of ischemia/ reperfusion (I/R) injury in experimental ovarian torsion in rats, on which low and high dose TNX was used during detorsion, in dose-dependent, histological, and biochemical analyses. METHODS: A total of 60 Wistar albino female subjects, which were 12-week old and weighed 260-305 grams (mean 277 grams), were used in the study. 8 different groups were formed, which were Group-1 (healthy, n = 4), Group2 (sham, n = 8), Group-3 (ischemia, n = 8), Group-4 (ischemia + TNX 10 mg/kg, n = 8), Group-5 (ischemia + TNX 20 mg/kg, n = 8), Group-6 (ischemia / reperfusion, n = 8), Group-7 (ischemia / reperfusion + TNX 10 mg/kg, n = 8), Group-8 (ischemia / reperfusion + TNX 20 mg/kg, n = 8). The ovaries of the anesthetized rats were reached with vascular clips. They were exposed to ischemia and reperfusion for 3 hours. In the ischemia-treated treatment groups, TNX intraperitoneal was given at the indicated doses 30 minutes before the ischemia. In the ischemia/ reperfusion treatment groups, TNX intraperitoneal was applied 30 minutes before reperfusion. Biochemical and histopathological examinations were performed from blood samples taken from their hearts and ovary tissues. RESULTS: Ovarian histopathological damage score (OHDS) and caspase-3 activities were increased in the ischemia group compared to the control and sham groups, while OHHS and caspase-3 levels were significantly lower in the I-10 and I-20 groups compared to the I group and in the I/R-10 and I/R-20 groups compared to the I/R group. We found MDA levels to be lower in the TNX treated groups than in the other groups, whereas GSH levels were increased in the TNX treated groups. There was a significant decrease in TNF-α in the I-10 and I/R-10 groups and in the I/R-10 group compared to the I/R group. CONCLUSION: The protective effect of tenoxicam in experimental ovarian I/R injury in rats was indicated by both histopathological and biochemical parameters.
