Abstract (EN)

Selcuk, C. (2017). Histomorphometric examination of the differences in the formation of new bone in the defects by administering systemic propolis to the rats in the critical defect model to be formed in the parietal bone in the calvarium of the rats. The aim of this experimental study is to determine the early and late new bone formation histomorphometrically by creating a critical size defect in the parietal bone in the calvarium of the intended rats and administering propolis via systemic gavage. In our experimental study, thirty rats were divided into six groups randomly. For the experiments each group has 5 rats were used in 6 groups of 30 Sprague-Dawley male rats ranging from 300-400 g. In the control groups, defects were formed in the calvarium of the rats and the defect was left empty and fed to the daily routine. B3 and B6 groups were left empty and 100 mg / kg systemic propolis was administered via gavage for 3 and 6 weeks. In the C3 and C6 groups, the defects of the rats were filled with alloplastic bone grafts and were also administered via a systemic propolis gavage at 100 mg / kg daily for 3 and 6 weeks. The first 3 groups of the experimental animals were sacrificed 21 days later and the second 3 groups were sacrificed 42 days later and the parietal bones were dis-carded. Histomorphometric examination was performed using a CCD digital camera (Optronics Microfire 1600x1200P, Goletaca, USA), a video card (ATI FireGL Advance Micro Device, Camberly, UK), a computerized motorized table (Heidenhein, Traunreut, Germany) and a light microscope (Leica DM 4000B, Wetzlar, Germany). Stereoinvestigator 7,5 (Microbrightfield, Willison, USA) program was used for the measurement. In addition, the Leica C Plan X10 lens (NA = 0.22) was used for volume measurements. As a result of the histomorphometric examination of the calvarias of the rats that were sacrificed for the 3 rd week comparing B3 and A3 group, it is found that there was a statistically significant difference in bone formation according to A3 group in B3 lgroup (p = 0.001). We also observed that there was a significant difference when comparing the A3 group and the C3 group to the new bone formation (p = 0.004). A statistically significant difference was not observed between group A6 and groups B6 and C6 when we performed a histomorphometric study of the rats we sacrificed for the 6th week ( Respectively: p = 0,762, p = 0,964). Our results 3 weeks to observe early bone formation, it is obtained a statistically significant difference between the control group (A3) and the graft group (C3) (p = 0.004). There was no statistically significant difference between the empty group (B3) and the grafted group (C3). (P = 0.981). According to the results of the rats sacrificed at the 6th week following late bone formation, There was no statistically significant difference between the control group (A6) and the graft group (C6) and between the null group (B6) and the graft group (C6). (In the order of p = 0.964, p = 0.994). As a result, we observed that the systemically administered propolis in this experimental model has a positive effect on early bone formation and has a statistically significant difference. However, no late effect on bone formation of administiration of systemic propolis was observed.
