The effects of grape seed ekstract on thioacetamide induced liver toxicity in rats

Abstract (EN)
Acute liver failure is a clinical condition stil with high mortality. Today, there are very few alternative treatment methods for acute hepatotoxicity. In this study we aimed to investigate the effects of grape seed extract (GSE) on thioacetamide (TAA) induced liver toxicity. In this study, 63 Wistar rats were divided into 9 groups. Group 1 was the control group. Group 2 was the sham group. Group 3: treated with low dose of GSE for 7 days (150 mg/ kg). Group 4: treated with high dose of GSE for 7 days (500 mg/ kg). Group 5: 100 mg/ kg TAA and simultaneous saline (SF) was administered for 2 days. In Group 6, low dose GSE was administered 3 days before 2 days TAA administration of 100 mg/ kg dose and application was also continued after TAA. In Group 7, high dose GSE was administered 3 days before 2 days TAA administration of 100 mg/ kg dose and application was also continued 7 days after TAA. In Group 8, low dose GSE was administered simultanously with 2 days TAA administration of 100 mg/ kg dose and continued for 7 days. In Group 9, high dose GSE was administered simultanously with 2 days TAA administration of 100 mg/ kg dose and continued for 7 days. In our study, two consecutive days of 100 mg/ kg TAA was given and liver toxicity was successfully induced. TAA administration was resulted with increase in proinflammatory cytokines such as nuclear Factor kappa B (NFk- B), Nuclear Factor erythroid-2-related factor 2 (Nrf2), Adenosine Mono Phosphate-Activated Protein Kinase (AMPK) and total oxidative stress (TOS) level and reduction in Total Antioxidant Level (TAL). High dose GSE administration was related with positive effects on AMPK levels and liver necrosis and inflammation parameters without significance. These findings suggest that new studies with high dose and long- time GSE treatment can provide important information on this subject.
