Investigation of the effects of taurine and apocynin on burn of stasi̇s zone in rats

Abstract (EN)
Aim: Burn trauma is one of the most severe traumas that a person can suffer. Care and treatment requires a special occupation and experience. Coagulation, stasis and hyperemia zones occur in the burned area of the skin. The central area where the burn is most severe is expressed as coagulation, the area around it as stasis, and at the outermost part, it is expressed as a zone of hyperemia. Tissues in the coagulation area cannot survive, and tissues inthe hyperemia zone often do not experience any pathological problems without the need for additional treatment. Especially in the stasis zone, a number of metabolic pathway start with the burn. Tissues in this area enter a critical period in which they can maintain their vitality by seeing the best response from treatment. Thsi metabolic process is related to how well the tissues in the stasis zone can overcome with oxidative stres. If the survival of the tissues in the stasis zone increases, further improvement is achieved in the burn area. In this study, we examined the effects of taurine amino acid and apocynin molecule on the survival of tissues stasiszone of the burned area. Material and Metod: The study was done withs rats weighing between 270 ± 20 gram. Burn areas were created by using a specially made metal brass comb of 55x19 mm in accordance with previously in the literature defined shape and stasis zones were obtained. Control, Burn, Taurine, Apocynin and Taurine+Apocynin groups were determined as 5 groups. The study was performed 21 days. Biopsies were taken for histological examination on the 3rd, 7th days and on the 21st days, that is, on the last day, biopsies were taken for biochemical evaluation and blood samples were taken for labaratory analysis. At he same time, photographs were taken for macroscopic evaluation on days 3rd, 7th and 21st. The rats were weighed at the begining and end of the experience. At the end of experience inflammation, re-epithelization, collagenization and CD34 immune reaciton were measured for histological evaluation. CAT, SOD, tGSH, TAS, TOS, MDA and PPC were measured for biochemical evaluation. In the labaratory examination, whole blood and blood biochemistry was examined. The weighings were recorded. The changes in the burned area were examined on the computer with the photographing. Results: In the examination performed histologically on the 3rd, 7th and 21st days inflammation was less on the 3rd day, in the other groups except the burn. Better reepithelization was observed in the Taurine group on the 7th day and significant reepithelization was observed in the other groups except the Burn group on the 21st day. In the biochemical evaluation, it was found that the effects of apocynin were excessive. Photographic examination showed significiant improvement in the Taurine+Apocynin group. There was no significant difference on weight. Taurine and apocynin had no toxic effects on liver and kidney. Conclusion: On the 3rd day, taurine and apocynin prevent inflammation. Taurine has more pronounced effects on the stasis zone in the early period (7th day), apocynin has a more pronounced effect on antioxidant enzymes and the late period (21st day). Taurine and apocynin create a synergistic effect, making the burn area more it was determined that it healed well. Key Words: apocynin, burn, burn healing, stasis zone, taurine
