Abstract (EN)

Circadian clock genes are the biological clock of the living thing and regulate the body's working mechanism. It consists of BMAL1, Period, Clock, Cryptochrome and Rev-Erbα genes. It shows an approximately 24-hour rhythm and affects the sleep-wake state, biochemical and psychological behavior of the living being. The aim of this study is to investigate the effects of prebiotic, probiotic and synbiotic treatment on circadian clock genes in the brain tissue of rats fed high fructose. In the study, 40 Wistar male albino rats were used and divided into 5 groups. For the first 7 weeks, they were fed with standard feed and water containing 20% fructose. Subsequently, additional treatment (probiotic, prebiotic, synbiotic) was applied for another 6 weeks. Euthanasia was performed without anesthesia, and 5cc of blood was taken from the neck of each rat, and then brain resection was performed. The mRNA expression levels of circadian clock genes (Bmal1, Clock, Rev-Erbα and Per2) in brain tissue were determined and compared with biochemical parameters. A strong positive correlation (p < 0.01) was found between CLOCK, BMAL1 and Per 2 genes, but no significance was found with the Rev-Erbα gene. Rev-Erbα gene showed a significant negative correlation with IL-6 and IL-1β, BMAL1 and Per2 genes showed a significant positive correlation with GSH and MDA elements (p < 0.05), CLOCK gene showed a significant positive correlation with GSH (p < 0.01). ) correlation was found to show a positive significant correlation (p < 0.05) with MDA.
