Researching of paraoxonase1 (PON1) Q192R and L55M gene polymorphism in patient with renal cell carcinoma

It is already known that increased oxidative stress has an effect on carcinogenesis. Oxidative stress has also an effect on the development of renal cell carcinoma (RCC). There are antioxidant systems operating against oxidative stres in the human body. One of these systems is paraoxonase enzym which protects low dansity lipoproteins (LDL) against oxidation. PON1 Q192R and L55M gene polymorphisms were researched in RCC patients in this study.Sixty patients with RCC and sixty healty control group were included in this study. Their peripheric blood samples were taken and DNA was isolated. Genotypes were found by polymerase chain reaction (PCR) and Alw I restriction enzyms were used for Q192R polymorphism and Hsp92II restriction enzyms were used L55M polymorphism. Chi-square and Fisher?s exact tests were used for analizing datas.Statistical significiantly difference was found in patients with RCC and control group in PON1 Q192R polymorphism (P=0.045). There was found significiantly difference in patients and control group in Q and R allel frequency (P=0.013). Q allel was higher in patients group more than in the control group. R allel was higher in control group more than in the patients group. Between control and patients group no statistical difference was found in L55M polymorphism (P=0.276). Additionaly there was no statistical difference in L and M allel frequency (P=0.069).At the end of this study Q allel is higher and R allel is lower in patients group than in the control group and the difference in PON1 Q192R polymorphism between patient with RCC and the control group was showed. This result shows that R allel can be a protective factor in RCC. According to our results there was no relation diagnosed between RCC and PON1 L55M polymorphism.
