The association of klotho gene polymorphism with life expectancy and cardiovascular events

Genetic variant of klotho have been reported to be associated with human longevity and atherosclerotic vascular events and risk factors. We aimed to evaluate the association between klotho gene polymorphism (KGP) and life expectancy and cardiovascular event in renal replacement therapy. We included that 51 chronic peritoneal dialysis (Group 1), 27 hemodialysis (Group 2), 25 renal transplant patients (Group 3) and 39 healthy subjects (Control Group 4). Assays for intact parathormon (iPTH), fibroblast growth factor 23 (FGF23), calcium, phosphorus, hemoglobin, HbA1c, 1-25(OH)2D3 and 25(OH)D3 were performed in patients and control groups. In our study, cardiovascular events and mortality states of the patient and control groups between January 2009 and December 2019 were recorded retrospectively. ɑ-Klotho gen mutations (functional polymorphism, F352V rs9536314) were evaluated by real time PCR and high resolution melting method. There was no significant difference between the groups in terms of KGP. There was no significant association between KGP and FGF23 levels. FGF23 has been shown to be an independent risk factor for mortality. There was no significant association between KGP and cardiovascular events, but patients with KGP wild were shown to have a higher risk of mortality. In conclusion; It was observed that the total mortality rate was higher and the survival rates were lower in the patient group with KGP wild.
