Demet Sumer "Effects of Resolvin D1 in Cerulein-Induced Acute Pancreatitis and Associated Lung Injury in Rats" Bulent Ecevit University Faculty of Medicine. Thesis in General Surgery, Zonguldak 2013. Introduction-Purpose: Acute pancreatitis (AP) is an inflammatory disease. The massive inflammatory reaction exacerbates damage by secretion of proinflammatory cytokines with subsequent release of proteases and reactive oxygen species. Subsequent entry of those into the systemic circulation always lead to multi-organ complications; especially lung injury. Resolvin D1 (RvD1); exerts potent anti-inflammatory, specifically, limit the actions of polymorphonuclear leukocytes and regulate leukocyte functions. The aim of this study was to examine the effect of RvD1 on AP and associated lung injury. Material and Methods: Rats (n=30) were randomly divided into 3 groups; 1) control group; 2) cerulein-induced AP group 3) cerulein-induced AP with RvD1 treatment group. Animals were given hourly intraperitoneal (ip) injections of normal saline or saline containing cerulein (50 μg/kg) for five h. RvD1 (100 ng/kg) was administred after and 24 h after last cerulein injections. Rats were sacrificed 24 hours after RvD1 injection, blood and tissues samples were harvested for biochemmical and histopathological analysis. Results: Pancreatitis was manifested by significant rise in plasma lipase activity in AP group. RvD1 treatment reduced increase in plasma activity of lipase. In AP group serum levels of TNF-α and IL-1β were significantly elevated, whereas administration of RvD1 reduced the increase. MDA levels and MPO activity was measured in lung and pancreatic tissue samples. In the AP group those were significantly higher compared to the control group. After RvD1 treatment MDA levels and the MPO activity were significantly lower in the RvD1 group than AP group. Pancreatic sections from cerulein-administered rats revealed edematous expansion of interacinar septae thus received significantly high scores. Treatment with RvD1 protected the pancreas from histological damage, as indicated by low histological scores. In AP group histological examination of lung sections revealed tissue damage characterized by edema and inflammatory cell infiltrate. RvD1 treatment resulted in reduction in lung injury scores.
