Investigaton of the antioxidant effect of resveratrol on human lung cancer cells (H1299)

Abstract (EN)
In this study, the antioxidant effects of resveratrol, which is a polyphenolic content, on human lung cancer cell line (H1299) were investigated. The antioxidant effect were displayed measurement of MDA and GSH level, and Se-GPx, non-Se-GPx, GST and GRx activities of the cells which were exposed to H2O2 which is a strong oxidant, after preincubated with resveratrol. The human embryonic lung fibroblast cells (MRC-5) were used as control cells. The cytotoxic effect of resveratrol and H2O2 on cells and the protective effect of resveratrol (antioxidant) against H2O2 cytotoxicity were measured by using the Cell Viability Test (Cell Titer-BlueR Cell viability test Kit). The effects of resveratrol on membrane were observed by fluorescence spectrophotometric measurement of the amount of MDA formed in the cells. Quantitative protein quantities were measured by the Bradford method. Glutathione level and activities of antioxidant enzymes were observed by spectrophotometric measurements. As a result of 24, 48 and 72 hours incubation of the cells with resveratrol, the values of IC50 were respectively calculated as 200 μM, 100 μM and 50 μM. IC50 and IC70 values were respectively found to be 300 μM and 420 μM after 72 hours of incubation of the cells with H2O2 which is a strong oxidant. Concentrations of resveratrol which are lower than IC50 showed protective (antioxidant) effect against H2O2 cytotoxicity in cells. The most effective dose was found as IC30. When the cells were incubated at IC50 and IC70 resveratrol concentrations, the amount of MDA increased. Consequently, this has shown that high concentrations of resveratrol cause membrane damage. The decreasing amount of MDA in the cells, which were exposed to H2O2 after pre-incubation with IC30 resveratrol, has shown that resveratrol has a membrane protective (antioxidant) effect against H2O2. Se-GPx, non-Se-GPx, GST and GRx activities of the cells exposed to resveratrol at IC50 and IC70 concentrations for 72 hours were found significantly higher than the control cells. Resveratrol has been shown to act as prooxidant at higher concentrations. These activities were decreased in the cells, which were exposed to H2O2 after pre-incubation with IC30 resveratrol. Resveratrol had oxidant and antioxidant effects on the cells and it was showed those effects were depending on concentration.
