Özet (EN)
Silver nanoparticles (Ag-NPs), which are used in many fields from health to cosmetics in the nano material industry, have been one of the fastest growing product categories. There are many studies reporting the toxicity effects of chemical synthesized NPs. Hence, during recent years, studies on biosynthesis of NPs have increased. By means of biosynthesis of Ag-NPs from some plant species which naturally contain strong reducing and stabilizing agents, it is aimed to develop solutions related to the deficiencies and some problems encountered in this are In this thesis, we have designed a simple and efficient green chemistry approach for the synthesis of colloidal silver nanoparticles that is formed by the reduction of silver nitrate (AgNO3) solution using P.kesrouanensis (Ayıgülü) and R. ribes (Işgın) leaf and stem extract (as fresh and sun dried). Ag-NPs were synthesized by using 1 mM AgNO3 solution and optimum conditions for nanoparticle synthesis were determined using UV-Vis spektrofotometer. FT-IR, TEM, SEM, EDX, XRD, DLS and Zeta Potential analyses were performed for the characterization of the Ag-NPs. The accumulation and elimination rate of Ag-NPs at different concentrations in A.salina was determined by ICP-MS analysis for 24, 48 and 72 hours periods. Accumulation increased due to time and concentration increase. The cytotoxic effect of biosynthesized silver nanoparticles on HUVECs cells was evaluated. The results showed that silver nanoparticles at concentrations below 100 µg/mL were biocompatible with the cell.
