Protective and therapeutic effect of rosmanol against oxidative stress-induced oxidative damage in gingival-derived fibroblast cells

Abstract (EN)

The aim of this in vitro study was to evaluate the protective and therapeutic effects of rosmanol administration on oxidative damage before and after hydrogen peroxide (H2O2)-induced oxidative damage in human gingival fibroblast (HGF) cells. IGF cell line was used in our study. IGF cells were induced with 200 μM H2O2 to induce oxidative stress in the cells. Before and after induction of oxidative stress, 4 different concentrations (0.1, 0.5, 1 and 5 μM) of rosmanol were applied and the effect on the viability of IGF cells was examined by XTT assay. Considering the data obtained from the study, it was determined that when rosmanol was applied to IGF cells before oxidative damage, it reduced cell damage by 2.5%-7.2%, and when applied after oxidative damage, it reduced cell damage by 3.2%-4.9%. No effect on cell viability was observed at 0.1, 0.5 and 1 µM rosmanol concentrations at doses where only rosmanol was applied to HGF cells. At 5 µM rosmanol concentration, cell viability was 94%. It was determined that rosmanol applied at concentrations of 10, 25, 50 and 100 µM decreased cell viability in a dose-dependent manner in HGF cells. In conclusion, rosmanol application was found to be protective and therapeutic in oxidative stress induced by H2O2 in HGF cells. The results obtained are promising for the use of rosmanol in the prevention and treatment of oxidative stress-induced inflammation and tissue damage such as periodontitis and further studies are needed.
