Terpenoids are compounds in the group of secondary metabolites produced by many organisms. Secondary metabolites can be produced in microorganisms by classical microbiological methods, as well as by metabolic engineering methods in recent years, they can be produced in commercial sizes. In this study, it was aimed to increase the amount of acetyl-CoA, which is the main component of the acetyl-CoA pathway, which is the production pathway of many terpenoids, by using S. cerevisiae as the background cell, by metabolic engineering methods. SCIGS22a strain originating from S. cerevisiae and used in the production of sequalene by metabolism engineering studies was used as a background strain and genetic modifications were made on this strain. On the pathway, deletion of the ypl062w distant region gene in the background cell was performed to prevent toxicity that may occur as a result of the production of terpenoids. In addition, a new support was provided to the formation of acetyl-CoA by increasing the backward activity of the ethanol production process, which reduces the formation of acetyl-CoA and shares acetaldehyde, the previous product of the pathway before acetyl-CoA.Cells formed during the modification and with those modification new strains for increasing acetyl-CoA amount made.
