Determination of heavy metal resistance and total protein of Saccharomyces cerevisiae 4 and Saccharomyces cerevisiae 2S1 TP(3-2)

Abstract (EN)

In this study, it is aimed that the effects of heavy metals on Saccharomyces cerevisiae 4 and Saccharomyces cerevisiae 2S1 TP (3-2) in [NiSO4, AgNO3, CuSO4.5H2O, CdSO4, ZnSO4.7H2O, CoSO4] and total protein quantities are determined. By using concentration values of metal salts between NiSO4, 100?1700 mg/L; AgNO3, 5?300 mg/L; CuSO4.5H2O, 500?2400 mg/L; CdSO4, 5?200 mg/L; ZnSO4.7H2O, 10?2000 mg/L; CoSO4, 100?1750 mg/L minimal inhibition concentration (MIK) levels and statistical probit values of strains aganist metals were determined. MIC values in the S. cerevisiae 4 were determined as AgNO3 for 200 mg/L and LC50 values 117,435 mg/L, CdSO4 for 100 mg/L and LC50 values 3,420 mg/L, CoSO4 for 1600 mg/L and LC50 values 717,102 mg/L, CuSO4.5H2O for 1700 mg/L and LC50 values 914,851 mg/L, NiSO4 for 1600 mg/L and LC50 values 566,554 mg/L, ZnSO4.7H2O for 1800 mg/L and LC50 values 339,215 mg/L respectively; also MIC values in the S. cerevisiae 2S1 TP (3-2) were determined as AgNO3 for 100 mg/L and LC50 values 52,480 mg/L, CdSO4 90 mg/L and LC50 values 0,057 mg/L, CoSO4 for 1250 mg/L and LC50 values 266,979 mg/L, CuSO4.5H2O for 2300 mg/L and LC50 values 1384,050 mg/L, NiSO4 for 1000 mg/L and LC50 values 624,241 mg/L, ZnSO4.7H2O for 1500 mg/L and LC50 values 394,156 mg/L respectively. Yeast strains grown in low and high heavy metal concentration and total protein quantities were determined comparing with control medium. It was found that the total protein quantities of yeast decreased with increasing heavy metal concentrations.
