Abstract (EN)
Objective:There is an active eNOS enzyme similar to endothelial cells in terms of both structure and NO production capacity in erythrocytes.It has been shown that the eNOS enzyme in erythrocytes is stimulated by ATP, it is not known whether this enzyme has a functional significance in the ATP-mediated vasodilation response.The aim of this study is to investigate whether the erythrocyte eNOS enzyme has a role in the vasodilation response in response to ATP. Method:Aortic rings obtained from 30 wistar rats were suspended in the organ bath. ATP-mediated vasodilation responses were examined in the presence of Krebs Solution or erythrocytes obtained from healthy individuals and pregnant women. NOS enzyme inhibitor L-NAME, eNOS substrate L-arginine was repeated in the presence of arginase enzyme inhibitor. 5 ml venous blood samples were taken from healthy and pregnant individuals participating in the study, intracellular NO formation and intracellular eNOS activity were evaluated. Results:It was shown for the first time that NO, produced by the erythrocyte eNOS enzyme, also plays a role in the ATP-mediated vasodilation response, along with NO from endothelium. We hope that our findings will provide a new contribution from the erythrocyte window to the ATP-mediated vasodilation mechanism, which has an important role in the local control of blood flow. On the other hand, our study revealed that erythrocyte eNOS enzyme activity was regulated differently in pregnancy conditions than in control conditions. Conclusion:It was shown for the first time that NO, produced by the erythrocyte eNOS enzyme, also plays a role in the ATP-mediated vasodilation response, along with NO from endothelium. We hope that our findings will provide a new contribution from the erythrocyte window to the ATP-mediated vasodilation mechanism, which has an important role in the local control of blood flow.
