Investigation of ubicuitin-proteasomy path proteins in healthy and diseased dental and implant environmental tissues

Abstract (EN)
Objective: Bacteria are the main factors in periodontitis and peri-implantitis, two of the inflammatory diseases of the mouth. Pathogenesis initiated by bacteria is accompanied by the inflammatory response of the host and the severity of the disease increases. Control of this inflammatory response is provided by the ubiquitin proteasome pathway (UPS), which is involved in many situations in the body. In the case of inflammatory disease, differences are seen in the levels of UPS proteins. In this study, it was aimed to determine the levels of UPS proteins in gingival and peri-implant crevicular fluid samples taken from individuals with periodontitis and peri-implantitis, by ELISA method compared to healthy individuals around teeth and implants. Method: In the study were included systemically healthy patients who were not pregnant or suspected, non-smokers and volunteered to participate. Two groups with healthy tooth and implant periphery were studied as the control group, and 2 groups with periodontitis and peri-implantitis were studied as the disease group. In the study, which consisted of 15 individuals and consisted of a total of 60 individuals, the levels of ubiquitin, E1, E2, E3, A20, UCHL1 which are UPS proteins in the gingival crevicular fluid and peri-implant crevicular fluid samples were examined by ELISA method. Results: When the E1 levels between the groups were evaluated, no statistically significant difference was found. Ubiquitin levels; It was found to be significantly higher in the peridontitis group compared to the healthy tooth group. E2 levels; It was found higher in peri-implantitis group than all other groups. When E3 and UCHL1 levels were evaluated, it was found to be significantly higher in the peri-implantitis group compared to the health tooth group. A20 levels were found to be significantly higher in the healthy tooth group than in all other groups. Conclusion: In this study, it was observed that the levels of UPS proteins (ubiquitin, E2, E3, UCHL1) involved in inflammation increased in inflammatory diseases such as periodontitis and peri-implantitis. A20 levels were higher in healthy tissues.
