Abstract (EN)

Pyometra in dogs is a disease frequently encountered in veterinary medicine and can result in death. The cystic endometrial hyperplasia (CEH)-pyometra complex was first described in 1957 and was explained as triggering the formation of pyometra because the uterus is sensitive to secondary infections as a result of hormonal changes. Studies investigating the prevalence of pyometra disease in dogs are quite limited. When the studies were examined, the prevalence of endometritis diagnosed during routine uterine examination was reported to be approximately 43%, and in the study conducted in our country, the prevalence of pyometra was reported to be 35%. These data have shown that the mortality rate of the disease is high throughout the world and the country and that existing methods for diagnosis and treatment must be taken to a more advanced level. Studies have shown that micro-ribonucleic acids (miRNAs) are associated with the diagnosis of many diseases, such as diabetes, atherosclerosis, kidney diseases, and cancer. The regulatory functions of miRNAs in the physiological state and pathological disorders of the normal uterus were evaluated, and animals with endometritis and a healthy uterus were examined in terms of miRNA expression profiles in the presence of uterine inflammation, and the presence of various candidate miRNAs was indicated. In our thesis study, we aimed to compare the expression levels of non-coding structures consisting of approximately 22 nucleotides, which play a role in epigenetic mechanisms such as miRNA, can show specific biomarker properties in the diagnosis of diseases, and are of great importance in targeted therapy, in uterine samples taken from healthy dogs and dogs diagnosed with pyometra. We concluded that miR-155 and miR-451, which were identified as candidates and can be specifically expressed in relevant tissues in cases of inflammation, are expressed more especially in the uterine tissues of animals with pyometra compared to healthy ones. In this context, with our thesis study, we would like to state that two miRNAs will make serious contributions to the early diagnosis of pyometra disease, which forms in the uterus and can be detected late. The results obtained from our study will provide a new perspective on the diagnosis and treatment of pyometra.
