Abstract (EN)

Macromolecular complexes-Biopolymere systems comprising a biocompatible synthetic polymer and a biomolecule targeting functional regulation are used in a wide range of applications including protected drugs, synthetic vaccines, diagnostic kits, biotechnological products, etc.In our thesis, we studied structural properties of biopolymer system, namely bioconjugate which developed as synthetic peptide antigens 40-60 amino acids long of VP1 capsid protein of Foot-and-Mouth Disease Virus (FMDV) bound spontaneously to polyelectrolyte carrier that shows an adjuvant effect.Bioconjugate synthesis was achieved with activation of polymer (PAA) with carbodiimide and with peptide binding. Using various physicochemical methods (HPLC, FTIR, Viscotek-with Ultra-viole Visible, Light Scattering, and Viscosity Quadruple Detector Systems, Fluorescence Spectroscopy), its structure and characterization, binding mechanism of the polymer with synthetic peptide, structure of bioconjugate particles and structural conversions due to reaction conditions were investigated.Synthesized bioconjugate was purified by passage through a Bio-Rad column. FTIR analysis was used for the purity of the product.Immunological properties of this conjugate were analyzed with ELISA method.Key words: Polyelectrolyte, Biopolymer systems, Immunogen, Polymer-Peptide conjugates, Foot-and-Mouth disease virus
