Abstract (EN)

Satureja spicigera (C. Koch) Boiss. (Trabzon Thyme) is an aromatic plant species belonging to the Lamiaceae family, and is widely distributed in the Eastern Black Sea region. As being evaluated as spices, flavorings and antimicrobial/antioxidant preservatives, this plant can be employed in food, pharmaceutical, cosmetic industries. Micropropagation potential of this plant was investigated by using basal media such as Murashige ve Skoog (MS, 1962), Linsmainer ve Skoog (LS,1965), Gamborg B5 (G B5, 1968) as well as plant growth regulators, such as giberellic acid (GA3), indole butyric acid (IBA), 2,4-dichloro phenoxy acetic acid (2,4-D), 6-benzyl amino purine (6-BA), 6-furfuryl amino purine (kinetin). The best gerimnation was observed from B5 basal medium supplemeted with 1 mg/L GA3, whilst the highest callus biomass was obtained from MS with 2 mg/L IBA plus 2 mg/L 6-BA, the most favorable shoot development in indirect organogenesis was from MS with 1 mg/L IBA plus 2 mg/L kinetin and finally the best somatic embryo yield was ontained from MS supplemented with 5 mg/L 2,4-D plus 0,1 mg/L kinetin. Seven different phenolics (gallic acid, protocatequic acid, protocataquic aldehyde, chlorogenic acid, benzoic acid, ferulic acid and rosmarinic acid) were analysed by HPLC both qualitatively and quantitatively in the extracts ontained from callus culture lines. The highest yield (42,3 ?g / g dry weight.). of essential oil was from those seedlings grown on the MS basal media supplemented with 1 mg / L 2,4-D and 2 mg / L kinetin.Key Words: Satureja spicigera, Microprogation, Phenolics, HPLC
