Investigation of Irisin Peptide in Carp (Cyprinus carpio ) and Rainbow Trout (Oncorhynchus mykiss) by ELISA method In this study, it was aimed to investigate the irisin peptide in carp (Cyprinus carpio) and rainbow trout (Oncorhynchus mykiss) by ELISA method. 80 fish in total, which are 40 C. carpio (20 seed fish+20 market sizes) and 40 O. mykiss (20 seed fish+20 market sizes), were used in this research. After anaesthetized fish using phenoxyethanol were subjected to length and weight measurement, the fishtails were cut off, the blood taken from caudal vein were centrifuged and the supernatants were stored at -20°C. The irisin peptide level measurement in blood serum of fish were carried out using an ELISA kit coded as MBS108944, produced by MyBioSource Fish Irisin (IS). The irisin peptide level difference between different genders of same species and between different species were determined by Independent-Samples t-test. The correlation analysis of gender, weight and total length in terms of irisin level in fish were investigated using Pearson Correlation Analysis. The value of P< 0,05 was considered statistically significant. The irisin peptide level difference between seed fish C. carpio-market size C. Carpio, male market size O. mykiss-female market size O. mykiss and male market size O. mykiss-male market size C. carpio were statistically significant (P< 0,05). The correlations of market size O. mykiss (p=0,033 r=0,477) and male O. mykiss (p=0,041 r=0,596) were determined regarding with total length. Consequently, irisin peptide was found in both C. carpio and O. mykiss; however, the differences were determined in terms of gender and total length. On the other hand, weight of the fish did not have a significant effect on irisin level statistically. The irisin level can differ by species and fish size; therefore, many studies must be carried out to generalize this scope of thought. Key Words: Irisin, C. carpio, O. mykiss, ELISA, Blood serum
