Effects of nesfatin-1 on contraction-relaxation mechanisms i̇n rat aorta

Nesfatin-1 is likely to be a new metabolic hormone recently discovered which is thought to have the significant endocrine and metabolic functions. The study carried out in the light of this data attempts to find the effect of Nesfatin-1 on the aorta contractions of 4-6 month-old male Wistar rats weighing 200–250 g. In this study the dose-dependent effect of nesfatin given to the aorta of rats at doses of 0.1, 1 ve 10 nM non-cumulatively methods on isometric contractions has been tested by using isolated organ bath. In addition, the effect of this application on aortic tissue of rats has been histologically presented using histo-chemical staining methods. It has been revealed at the end of the study that nesfatin-1 at different doses inhibits the contractions of rat aorta in a dose dependent manner and this application results in immunohistochemical changes in rat aorta. In terms of the contraction and relaxation stress percentages it has not resulted in any statistical difference within the group at a dose of 0.1nM nesfatin while there is a statistically significant difference among the groups at doses of 1 nM and 10 nM(p<0.05). Nesfatin application at doses of 1 and 10 nM has been found to decrease the aortic contractions in a statistically significant way (p<0.05 ). The se results have demonstrated that nesfatin-1 inhibits both the aortic contractions and relaxation procedures of rats in a dose dependent manner. Moreover, these dose-dependent effects of nesfatin application have been histologically identified in tissue level, as well.
