The immunuhistochemical research of nerve growth factors involving pre and postnatal periods in rat's adrenal medulla

Abstract (EN)

In this study, the prenatal and postnatal development of the rat adrenal medulla cells is aimed to be investigated through using original antibodies with immunohystochemical methods at the level of light microscope. For this reason, adrenal tissue samples were taken from rats of age ranging from embryonic 16th and 19th days, newborn, young, adolescent and elderly and gone through the conventional light microscope check and paraffin blocks were prepared. Indirect immunohystochemical methods as well as preparates which were prepared by using FGF, NGF and NF antibodies were analyzed in Leica DM 4000 photo light microscope in order for the investigation of the medulla development under the scope of our study. It was detected that while FGF and NGF eclipses were being observed in ganglion and chromaffin cells in embryonic 16th day, NF eclipse was just only observed in sympathetic ganglion cells at minimum levels. While the FGF and NGF immune reactivity was being observed at similar levels in sympathetic ganglion and chromaffin cells in embryonic 19th day, the NF eclipse was observed as meager. In the period of newborn, FGF and NGF were detected as strong in the sympathetic ganglion cells during the immune painting done within FGF, NGF and NF antibodies, even though chromaffin cell eclipse was detected at minimum levels. It was seen that the negative eclipse observed against each three antibodies did not change with the progress of the development as well as the eclipses in sympathetic ganglion cells was observed to increase parallel to the age concerning the age groups starting from adolescence. Besides, the fact that medulla connective tissues increased during the adolescence was also seen as an interesting factor. As a result of our study, it was finally that while the chromaffin cell eclipses decrease parallel to the age and then show negative reaction, the sympathetic ganglion cell eclipses continue increasingly observed in the each of three antibodies concerning the periods investigated. Under the light of the obtained findings, FGF, NGF and NF were decided to be appropriate markers as neuronal determinants in definition of the sympathetic ganglion and chromaffin cell developments.
